All major airlines offer fast nonstop and direct service to Miami, Orlando and Tampa. Comair, Gulfstream all have direct flight from Miami, Orlando & Tampa/Naples to Key West.
You Take the Florida Turnpike south, exit 4 will connect you to the Turnpike extension which meets US 1 in Florida City. If your comming from Miami International Airport take LeJeune Road south to 836 West to the Turnpike Extension which flows into US1 at Florida City, the only way into the Keys. It's known as the Overseas Highway it takes about 3 hours once you have reached Florida City. I highly recomend taking this part of the trip by daylight since the views are magnificant. I have traveled this route for over 50 years and I've never ceased to be enthralled by the varied hues and shades of the sea.
When you enter the keys you will notice small green signs with white numbers--mile markers--on the right shoulder of the road. MM Zero is in the heart of Key West.

Greyhound services the Keys the Miami to Key West route has scheduled stops in Homestead, Marathon, Ramrod Key and Key West.
